Essential role of histidine peptides in tetanus toxin production.

چکیده
A number of papers from this laboratory have traced the development of a medium for the production of highly potent tetanus toxin, the basis of which is a pancreatic digest of casein (l-3). Considerable work has been done in an effort to identify those components of the digest essential for toxin production, and it has been shown that at least some of these materials are peptides (4, 5). In the present communication, experiments are reported which resulted in the identification of one such group of peptides.

Peptide and keto acid utilization in replacing phenylalanine in adaptive enzyme synthesis.
Glycylasparagine has been found to be more effective than aspartic acid in reversing the inhibition of adaptive enzyme synthesis by cysteic acid or P-hydroxyaspartic acid or combinations of these synergistic inhibitors (1). The results were interpreted to indicate a pathway of utilization of the peptide not involving the inhibited enzymatic utilization of the free amino acid. Also, histidine pe...
متن کاملExploring the functional domain and the target of the tetanus toxin light chain in neurohypophysial terminals.
The tetanus toxin light chain blocks calcium induced vasopressin release from neurohypophysial nerve terminals. Here we show that histidine residue 233 within the putative zinc binding motif of the tetanus toxin light chain is essential for the inhibition of exocytosis, in the rat. The zinc chelating agent dipicolinic acid as well as captopril, an inhibitor of zinc-dependent peptidases, counter...
متن کاملFunctional characterization of the catalytic site of the tetanus toxin light chain using permeabilized adrenal chromaffin cells.
The molecular events underlying the inhibition of exocytosis by tetanus toxin were investigated in permeabilized adrenal chromaffin cells. We found that replacement of amino acid residues within the putative zinc binding domain of the tetanus toxin light chain such as of histidine (position 233) by cysteine or valine, or of glutamate (position 234) by glutamine completely abolished the effect o...
متن کاملComparative Study of Three Vaccinal Strains of Clostridium tetani Including Harvard 52, G5 and 49205 from Standpoint of Six Essential Factors to Evaluate Their Toxigenesis for Use in Tetanus Vaccine Production
Tetanus is an important disease which created by tetanospasmin toxin of Clostridium tetani. In this study we surveyed six important factors including LF, KF, MLD, pH, OD and total protein assay of Harvard 52 (H52), G5 (HG5) and 49205 (H49205) strains of the bacterium to determine which of them were more suitable for use in vaccine production. The mentioned strains were seperatedly reactivated i...
متن کاملCloning and expression of tetanus toxin C fragment (Fc) in prokaryotic vector for constructing recombinant protein based vaccine for tetanus
Tetanus is a disease caused by tetanus toxin, a potent inhibitor for the release of inhibitory neurotransmitter in the central nervous system that causes spastic paralysis. Fragment C (52 kD) of this toxin is responsible for binding to the neuronal membrane. For this reason, and also its non toxigenic and immunogenic nature, this fragment might be ideal for new vaccine development. Presently, w...
